Keywords: PBS, RNase, RNase-free

Describes the preparation of RNase free PBS.

Whether the PBS is RNase-free or not depends on the grade of water used and the protocol can of course be adapted to suite ones needs.

Instructions

  1. Procedure
    8.0 gNaCl
    0.2 gKCl
    1.44 gNa2PO4
    0.24 gKH2PO4
    ? 1 literDEPC-H2O

Dissolve NaCl, KCl, Na2PO4 and KH2PO4 in 800 ml of DEPC-H2O. Adjust the pH to 7.4 using HCl.

To avoid contamination by RNase this is preferably done by dropping the solution on a pH strip.

Adjust the volume to 1 liter with DEPC-H2O. Sterilize by autoclaving.
