Describes the preparation of RNase free PBS.
Whether the PBS is RNase-free or not depends on the grade of water used and the protocol can of course be adapted to suite ones needs.
Required Materials
- NaCl
- KCl
- Disodium phosphate
- Potassium dihydrogenphosphate
- Sterile water , of DEPC quality.

Procedure
8.0 g NaCl 0.2 g KCl 1.44 g Na2PO4 0.24 g KH2PO4 ? 1 liter DEPC-H2O Dissolve NaCl, KCl, Na2PO4 and KH2PO4 in 800 ml of DEPC-H2O. Adjust the pH to 7.4 using HCl.
To avoid contamination by RNase this is preferably done by dropping the solution on a pH strip.
Adjust the volume to 1 liter with DEPC-H2O. Sterilize by autoclaving.
