Description of how to prepare a 5 mg/ml MTT solution

To be used in MTT cell viability assay based on the cleavage of the yellow tetrazolium salt MTT to purple formazan crystals by metabolic active cells. The formazan crystals formed are solubilized and the resulting colored solution is quantified by using a scanning multiwell spectrophotometer.

Prepare MTT solution in laminar air flow (LAF) bench.

    • Add 100 mL PBS buffer to a cup, place on magnetic stirrer
    • Add 0.5 g MTT powder

    Wrap cup in tin foil to protect from light, stir for 1-2 hours

    Sterile filter solution, split into aliquots of, for example, 10 ml and store at -20 °C
